Western blot analysis of diluted HeLa cell lysates using anti-β-Actin antibody. Lane 1 contained 2μg of total protein, and four 2-fold serial dilutions were applied to lanes 2-5 at 10μL/well.


Procedure Summary

1.       Wash membrane-bound HRP (antibody conjugate) three times for 5 minutes in TBS/T.

2.       Prepare 1X ECL Western Blotting Substrate by mix Reagent A and Reagent B at a 1000:5 ratio (i.e., 5mL Reagent A + 25μL Reagent B for 6 × 8 cm2membrane) and add it to the blot.

3.       Incubate Reagent with membrane for 1 minute, remove excess solution (membrane remains wet), wrap in plastic.

4.       Expose blot to X-ray film.
